Role of protein Ubiquitylation in tumor-cell invasion and migration

It has been demonstrated that blocking dissemination of tumor cells to distant sites could drastically reduce the mortality of patients with metastatic tumors, which represent the major cause of death from cancer. The Ubiquitin-Proteasome System (UPS) is a fundamental regulator of cellular protein homeostasis with high therapeutic potential in cancer treatment. The aim of my project is to contribute to the identification and characterization of new putative therapeutic targets within the UPS that regulate tumor cell migration and invasion. To this end, a shRNA genetic screen using a pooled library was performed, followed by the selection of a non-migratory resulting population. The identification of the candidate regulatory genes after the selection process is the next step in my project, which I intend to pursue during my visit to the laboratory of Dr Espinosa. He is the director of a sequencing facility, and his expertise will let me deconvolute the population of shRNA obtained after the selection process. The identification of the shRNA will let me choose the candidate genes for further study. This stage is not only crucial for the development of my experimental work but will significantly contribute to my scientific training in Functional Genomics.
